STRUCTURE AND FUNCTION
The metabolic role of Alpha Lipoic Acid as a cofactor in the multi-enzyme
oxidative decarboxylation complexes of the Krebs citric acid cycle has been
established for decades. The Krebs cycle is a major biochemical process that
derives energy from carbohydrates. Pyruvate dehydrogenase and a-ketogluterate
dehydrogenase are two central enzyme complexes that require Alpha Lipoic Acid
as a coenzyme for glucose metabolism and the Krebs cycle. Pyruvate
dehydrogenase complex (PDC) converts pyruvate to acetyl-CoA, one of the two
essential substrates needed to produce citrate. A deficiency in this enzymatic
complex may limit the production of citrate. Because citrate is the first
substrate in the Krebs cycle, the process cannot proceed. Alternate metabolic
pathways may be stimulated in an attempt to produce acetyl-CoA; however, an
energy deficit remains, especially in the central nervous system.*
More recently, Alpha Lipoic Acid has received attention for its role as a "universal antioxidant." Possessing the ability to neutralize free radicals in both the aqueous and lipid phases of the cell, Alpha Lipoic Acid scavenges hydroxyl radicals, hydrogen peroxide, peroxynitrite, hypochlorous acid, singlet oxygen and nitric oxide. SCIENTIFIC RESEARCH
- In order to investigate the antioxidant support of Alpha Lipoic Acid, Marangon et al., conducted study in which plasma, cholesterol and urine oxidizability were measured before and after 4 months of 600 mg/day Alpha Lipoic Acid supplementation. Compared to base line samples, Alpha Lipoic Acid significantly promoted the lag time of oxidation of plasma and cholesterol oxidation, and supported fewer isoprostanes in the urine, markers of oxidative stress.*
- Jacob et al., evaluated the effects of 600 mg Alpha Lipoic Acid in a placebo-controlled double-blind study of 74 subjects. All subjects were comparable at base line. Following 4 weeks of supplementation, significantly more subjects experienced an increase in sensitivity to sugar-metabolizing hormone when compared to placebo, as measured by an isoglycemic glucose-clamp.*
